ContraindicationsAdministration to dogs exhibiting previous hypersensitivity to carprofen.In the absence of specific data, administration to the cat is contraindicated. The elimination time of NSAIDs, including carprofen, is longer in the cat than in the dog, and the therapeutic index is narrower.
ActionsCarprofen is a non-narcotic, nonsteroidal, anti-inflammatory drug with potent analgesic and antipyretic activity. Carprofen is a reversible inhibitor of cyclooxygenase and a moderate inhibitor of phospholipase A2. It has a minimal effect on those prostaglandins that are necessary for the integrity of gastrointestinal tract and renal functions. In an in vitro study using canine cell cultures, carprofen demonstrated a greater than 100-fold selective inhibition of COX-2 compared with COX-1. Preliminary studies indicate that carprofen increases production of glycosaminoglycan in chondrocytes, thus slowing the process of cartilage loss.
PrecautionsAs a class, cyclooxygenase inhibitory NSAIDs may be associated with gastrointestinal and renal toxicity. Effects may result from decreased prostaglandin production and inhibition of the enzyme cyclooxygenase, which is responsible for the formation of prostaglandins from arachidonic acid. NSAID therapy could unmask occult disease, which has previously been undiagnosed due to the absence of clinical signs.Use with caution in dogs that are less than 6 weeks of age, debilitated and aged, with existing cardiac, hepatic or renal disease, dehydrated, hypovolaemic or hypotensive.Safety has not been established in dogs with bleeding disorders, pregnant or lactating bitches.Drug compatibility should be monitored closely in patients requiring additional therapy. Concurrent use of Rimadyl with other anti-inflammatory drugs such as corticosteroids and other NSAIDs, should be avoided.Due to the palatable nature of Rimadyl Chewable Tablets, store out of reach of dogs in a secured location. Adverse reactions may occur if large quantities of tablets are ingested.All dogs should undergo a thorough clinical examination and appropriate laboratory tests before introduction of NSAID therapy. During extended administration, appropriate re-evaluation and laboratory tests should be undertaken periodically.
Dosage and Administration Dogs. 2 to 4 mg/kg bodyweight/day. Dose, frequency and duration of treatment will be dependent on clinical response of the condition under treatment. Initial therapy at 4 mg/kg bodyweight/day given in two equally divided doses is generally recommended. Subject to clinical response, the dose may be reduced to 2 mg/kg bodyweight/day administered as a once daily dose.
