|Indication||Treatment and prevention of gastric ulcers in horses. Clinical signs of gastric ulceration include inappetence and weight loss, colic, dull appearance of the coat, crib biting, loss of competitive spirit and bad temperament.|
|Actions||Hydrochloric acid, secreted from parietal cells within the gastric mucosa of the equine stomach, is the primary aggressive factor in gastric ulcer disease. Hydrochloric acid is secreted continuously in the stomach and the rate of secretion is stimulated by feed intake and stress. The principal strategy of ulcer therapy is to administer treatment that will inhibit gastric acid secretion to maintain intragastric acidity above a pH of 4, thereby rendering the gastric fluid nonaggressive to the squamous mucosal epithelium. By maintaining a gastric pH of 4 or above, gastric lesions heal spontaneously and should not recur while treatment is maintained.Omeprazole is a dose dependent inhibitor of gastric acid secretion. Omeprazole regulates gastric acid secretion at the gastric parietal cell membrane by inhibiting the activity of the enzyme H+K+ATPase, which catalyses the exchange of hydrogen ions and is referred to as the proton pump. Omeprazole regulates the final step in hydrogen ion production, thereby preventing gastric acid secretion regardless of stimuli.Omeprazole has a long duration of action, which allows daily dosing. The antisecretory effect increases with repeated daily dosing and reaches a plateau after four days. Omeprazole readily accumulates in the acidic compartments of the gastric parietal cell with repeated dosing to provide an antisecretory effect that lasts up to 72 hours.|

|Dosage and Administration||Give orally via syringe. Foals. 2 mL/100 kg bodyweight (1 mg/kg) once daily as directed by a veterinarian. Adult horses. Treatment: 10 mL/500 kg (1 mg/kg) once daily for two weeks followed by daily maintenance dosing. Maintenance: 5 mL/500 kg (0.5 mg/kg) once daily while horses remain in training, as directed by a veterinarian.Gastrozol Daily 10 mL/500 kg can provide rapid healing of stomach ulcers while horses remain in training, which may be associated with the improvement in clinical signs within two weeks of commencing treatment.When left untreated, ulcers can recur within two weeks with a severity and across an area equivalent to that prior to treatment. Available data indicates that regular daily maintenance dosing at a reduced rate prevents the recurrence of ulcers.|
